摘要:研究大豆异黄酮(soy isoflavone,SIF)对雄性生殖细胞凋亡的影响。方法:成年组:SD大鼠随机分为五组,每组9只,孕鼠随机分对照组(玉米油)、低剂量组[SIF 50mg/(kg·bw·d)]、中剂量组[SIF 200mg/(kg·bw·d)]、高剂量组[SIF 400mg/(kg·bw·d)]和乙烯雌酚(diethylstilbestrol,DES)0.1mg/(kg·bw·d)组;从妊娠0天开始灌胃给予大豆异黄酮(SIF),子鼠21天断奶后开始灌胃大豆异黄酮(SIF)。哺乳期结束后子代雄鼠按照原组别重新分笼,每组9只,持续灌胃SIF直至90d试验结束。子代雄鼠在90天龄处死,观察各组子代雄鼠生殖系统发育相关指标,进行各项指标测定。结果:①49-90天龄子鼠大豆异黄酮200mg/(kg·bw·d)、400mg/(kg·bw·d)组以及0.1mg/(kg·bw·d)DES组的食物利用率显著下降(P<0.01)。②大豆异黄酮剂量为50mg/(kg·bw·d)级以上时,体重增长显著缓慢,不同年龄段的子鼠体重均显著下降(P<0.05)。③随着大豆异黄酮剂量的增加,90天龄子鼠指标的变化明显,睾丸和附睾的重量及睾丸脏器系数均显著降低(P<0.01)。④200mg/(kg·bw·d)大豆异黄酮组、400mg/(kg·bw·d)大豆异黄酮组以及0.1mg/(kg·bw·d)乙烯雌酚对照组的大鼠早期凋亡精子的凋亡率与玉米油空白组的大鼠早期凋亡精子的凋亡率之间有极显著性差异(P<0.01)。结论:长期接触大豆异黄酮对雄性大鼠生殖细胞有一定的损伤作用。

Abstract:To investigate soy isoflavones (soy isoflavone, SIF)the influence of male rats germ cell apoptosis.Methods: Sprague-Dawley male rats were randomly divided into five groups, nine rats per group. SIF were classed as control group (corn oil), low-dose group [SIF 50mg/(kg·bw·d)], middle-dose group [SIF 200mg/(kg·bw·d)],high-dose group [SIF 400mg/(kg·bw·d)], positive control group [DES 0.1mg/(kg·bw·d)]; Pregnancy rats receive orally from pregnancy day 0. After separating sucklings from their mothers on the 21st day, male offspring receive the same dose until reaching the age of sexual maturity. According to original group after weaning period, nine male rats per group, and then were fed until 90d. We observe the related indicators of reproductive system development. Body weight, organ weights and other parameters were measured after dissecting the rats. Results: ①the Food efficiency ratio for 200mg/(kg·bw·d), 400mg/(kg·bw·d) and 0.1mg/(kg·bw·d),DES declines significantly(P<0.05) from 49th to 90th day. ②when the dose of SIF surpasses 50mg/(kg·bw·d), male rats’ body weight increases slowly,the body weights of rats at various ages begin to fall significantly lower (P<0.05).③Along with the increase of SIF dose, testis weights and epididymis weights and the organ coefficient of the testis was significantly decreased (P<0.01).④With 200mg/(kg·bw·d), 400mg/(kg·bw·d))increase of SIF dose and 0.1mg/(kg·bw·d)DES dose,compared with the control group, a significant differences in the percentage of live and apoptotic sperm. (P<0.01). Conclusion: There existed toxicology effects of germ cell after long term soy SIF exposure to male rats.
