摘要:目的 探究老年痴呆症口腔微生物菌群结构的变化,为其预防老年痴呆症状奠定理论基础。方法:采用酚一氯仿方法提取样本茵群基因组DNA,并进行16S rRNA v3区片段PCR扩增,扩增产物用变性梯度凝胶电泳法(denaturing gradient gel electrophoresis,DGGE)检测分析,通过PCR扩增及DGGE电泳研究其健康小鼠和患有老年痴呆模式小鼠的对比。结果成功扩增了口腔菌群基因组DNA为230bp。DGGE电泳结果发现,DGGE分子指纹图谱上条带存在显著的差异,用bionumberics软件分析DGGE结果,健康小鼠腔菌群相似度高,而老年痴呆模式小鼠口腔菌群相似度低且健康小鼠与患病的相似度有明显的差异,可作为早期诊断及治疗的依据。

Abstract:[Objective] To study the oral microbial community structure of AD and lay a foundation for AD prevention . Methods: by phenolic chloroform extracted from floras genome DNA of samples, 16S rRNA V3 region PCR products were detected and analyzed by denaturing gradient gel electrophoresis (DGGE) to compare with the healthy and AD mice models. It is get 230bp floras genomic DNA and result shows that molecular fingerprint on the strip notable differences by DGGE analysis software, by bionumberics software analyzed oral microbial community high similarity degree of healthy, However, the oral microbial community of suffering AD mice models have low similarity, Oral microbial community of AD mice models is different from healthy ones, which can provide evidence for the early diagnosis and treatment.
