摘 要:面对纤维素物质利用率偏低的现状,有效降解纤维素资源,提高纤维素酶活和降低生产成本是当前亟待解决的重大问题。本实验从蝎子肠胃中分离得到13株放线菌,选择一株透明圈较大的菌株作为实验菌株,命名为九号菌株。为了提高该菌株的产酶活性,本实验对九号菌株产纤维素酶的时间、pH以及酶促反应温度进行了优化,通过优化,九号菌株的最适生长天数为5天,培养最适pH为6.2,其最适酶促温度为50 ℃。结果表明,通过改变培养条件以及掌握菌所产纤维素酶的酶学性质,酶活会大大提高,产酶效果也更佳。

Abstract:Facing with the low utilization rate of cellulose, effective degradation of cellulose resource, higher activity of enzyme and lower production costs are the current major problems to be solved urgently. In this experiment, thirteen actinomyces strain were screened from the stomach of scorpion. The one which has a big clear zone was selected as the experimental one, named the No.9 strain. In order to improve the activity of enzyme, the cultivation time, initial pH and temperature on enzyme catalytic reaction were optimized. Through optimization, the optimal cultivation time was 5 days, the optimal initial pH was 6.2, and its optimal enzyme catalytic temperature was 50 degrees centigrade. The results showed that by changing the cultivation conditions and grasping the characteristic of strain’s producing cellulase, the activity of enzyme would be greatly enhanced, enzyme production effects were also better.
