摘 要:芯片实验室(Lab on a chip)技术已发展成为当今世界上最前沿的科技领域之一,其以微流控芯片为核心,因高效快速、通量集成、节能低耗等特点在化学、生物学等领域的应用中彰显出独特的优势。多肽作为重要的内源性物质,在生物体内发挥重要的生理功能,多肽类药物、疫苗以及诊断试剂已成为生物医药领域的研究热点。本论文将多肽筛选与微流控芯片技术相结合,充分发挥微流控芯片的优势,构建了微流控芯片的多肽筛选识别新系统,开展了基于微流控芯片的多肽筛选与检测研究。主要研究结果如下:
选取生理活性多肽特定位点的氨基酸,进行组合化学肽库合成,采用混合-裂分法构建了“一珠一物”多肽文库,库容量为105;将靶标蛋白与文库中的多肽微珠相互作用,采用生物素-链霉亲和素桥连及磁珠捕获的方法对多肽微珠进行特异性筛选;相对于传统的基于光学方法检测的多肽文库筛选,该方法具有高效、简便特点,又由于其无假阴性,可将大容量的肽库进行简并;初步将该筛选体系应用与于微流控系统,构建自动化、高效率、高通量的组合化学多肽筛选平台。
