摘要:为分析斑玉蕈提取物的抗氧化活性,采用乙醇浸提斑玉蕈得乙醇粗提物(ECE),ECE分别通过正己烷、乙酸乙酯、正丁醇萃取,得到正己烷萃取物(HF)、乙酸乙酯萃取物(EAF)、正丁醇萃取物(BF)和水提物(WF);以清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基能力、清除羟基自由基能力、还原能力、螯合金属离子、清除超氧阴离子能力为指标,评价各种溶剂提取物的抗氧化活性,并测定了各种提取物的总酚含量。结果表明:斑玉蕈提取物的各个组分清除DPPH自由基能力、清除羟基自由基能力、还原能力、螯合金属离子、清除超氧阴离子能力均表现出一定的浓度依赖性;EAF对DPPH自由基的清除活性最高,其半数有效浓度(EC50)值为0.19 g/L,略低于阳性对照维生素C(VC);EAF对铁离子的螯合能力最强,EC50值为2.40 g/L;EAF和ECE清除超氧阴离子的能力要高于VC;EAF[(35.20±0.12) mg/g]的总酚含量最高;总酚含量与斑玉蕈提取物清除DPPH自由基能力、还原能力、清除羟基自由基能力、清除超氧阴离子能力均呈显著正相关(P<0.05)。

Abstract:Hypsizygus marmoreus were extracted by ethanol, which were further extracted by hexane, ethyl acetate and butanol respectively. The antioxidant activities of ethanolic crude extract (ECE) and four solvent sub-fractions [namely, hexane fraction (HF), ethyl acetate fraction (EAF), butanol fraction(BF) and water fraction(WF)] were investigated using five different in vitro antioxidant assays, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities, reducing power, hydroxyl radical scavenging activities, chelating ability, superoxide anion scavenging activities. Total phenols(TP) content of Hypsizygus marmoreus extracts were determinated. The results showed that DPPH radical scavenging activities, reducing power, hydroxyl radical scavenging activities, chelating ability and superoxide anion scavenging activities of the Hypsizygus marmoreus extracts were dependent on their concentrations. EAF showed the most potent capacity for scavenging DPPH with half maximal effective concentration (EC50) values reaching 0.19 g/L, which was slightly lower than Vitamin C (VC). EAF exhibited the highest capacity for ferrous ions chelating activities with EC50 values reaching 2.40 g/L. The capacity scavenging of EAF and ECE were better than VC. TP content in EAF was (35.20±0.12) mg/g. Thus DPPH radical scavenging activities, reducing power, hydroxyl radical scavenging activities and superoxide anion scavenging activities of Hypsizygus marmoreus seem to be positively correlated with TP content (P<0.05).
