摘要:建立GC内标法测定留兰香精油中萜类成分含量的方法。方法:采用制备薄层色谱法分离纯化其中的萜类成分获得对照品,并用核磁波谱法进行结构鉴定,用GC内标法进行含量测定。结果:从留兰香精油中分离纯化得到2个化合物,分别鉴定为香芹酮和柠檬烯,完成了对其的含量测定,建立了GC内标法测定留兰香中萜类成分含量的方法。香芹酮在0.3mg/ml-1.8mg/ml线性良好,Y=1.6486X-0.1442,r =0.9996.回收率为98.76%,RSD为1.53%;柠檬烯在0.1mg/ml-1.5mg/ml线性良好,Y=1.7808X-0.0451,r =0.9996,回收率为97.24%,RSD为1.59%.结论:采用制备薄层法分离留兰香精油中的主要化学成分方法简便,分离物纯度高。GC内标法进行含量测定准确可靠,重现性好。

Abstract: Objective To establish a method for determining the content of terpenoids in Mentha spicata L by GC internal standard method. Methods Adopt the preparative thin layer chromatography to separate and purificate the terpenoids components and obtain the reference substances .Identify the structures by NMR and determine the content by GC internal standard method. Results Get two compounds that can be as reference substances and identify their structures,carvone and limonene.Complete their content determinations and establishe a method for determining the content of terpenoids components in Mentha spicata L by GC internal standard method.The linearity of the carvone in 0.3 mg/ml - 1.8 mg/ml is good,y=1.6486x- 0.1442,r=0.9996.The recovery rate is 98.76%, RSD= 1.53%.The linearity of the limonene in 0.1 mg/ml - 1.5 mg/ml is good, y=1.7808x-0.0451,r=0.9996.The recovery rate is 97.24%, RSD=1.59%. Conclusion Adopting the preparative thin layer chromatography to separate and purificate the terpenoids components is simple, the purity of the compounds is high.GC internal standard method for content determination is accurate and reliable.The repeatability is also good.

本课题研究采用制备薄层法分离留兰香精油中的主要萜类成分,此方法简便,分离物纯度高。GC内标法进行留兰香中萜类成分含量测定方法简单,准确可靠,重现性好,所测结果对留兰香的生产应用将起到一定的指导作用,对其他挥发油的含量测定也将起到一定的借鉴作用。
